10 Ta Estimation of the melting and annealing temperatures of primer: If the primer is shorter than 25 nucleotides, the approx. melting temperature (Tm) is calculated using the following formula: Tm= 4 (G + C) + 2 (A + T) G, C, A, T - number of respective nucleotides in the primer. Annealing temperature should be approx. 5°C lower than the melting temperature.

12 Dimethyl Sulfoxide (DMSO) solution
DMSO has been shown to facilitate DNA strand separation (in GC rich difficult secondary structures) because it disrupts base pairing and has been shown to improve PCR efficiency.

13 Formamide solution Formamide has been shown to decrease the temperature necessary for the reassociation of complementary nucleic acids and has been shown to improve PCR efficiency, particularly on difficult secondary structures.

14 Betaine solution Betaine has been shown to improve the amplification of DNA by reducing the formation of secondary structure in GC-rich regions. It is an isostabilizing agent, equalizing the contribution of GC and AT base pairing to the stability of the DNA duplex.

15 Tetramethylammonium chloride (TMA)
Zvyšuje teplotu annealingu primerů, potlačuje vnik nespecifických produktů. Váže se na AT bohaté oblasti a zamezuje jejich přednostnímu tání v porovnání s CG bohatými oblastmi.

Stabilizuje enzymy a zabraňuje jejich adhezi na povrch zkumavek.
